Yes remove plaster so that your new studs can fix directly to the old existing plates and noggins, you will also need some type of head over the door assuming its a 2040mm. assuming 820mm door allow 850mm between your new studs.
hi guys I want to build a door frame between two walls the opening is about a metre I have found 3 noggins top centre and bottom do I need to screw in a stud and then the door jamb and do I need to remove the plaster or just put it over it. thx![]()

850 is not enough between studs, 820 door plus jambs 38 mm plus margin each side 6mm is 864 tight . Leave about 10 mm for packing plumb and you should have 875 between studs and for a 2040 door head height should be around 2100.
I wouldn't cut out plaster if you don't have to as once cut the plaster will need extra support between noggin and plates.
